ABSTRACT

Psoraleae Fructus (PF) is a non-toxic Chinese herbal medicine, while the liver injury caused by PF has aroused wide concern in recent years. At present, animal experiments and in vitro studies have been carried out to explore the mechanism, targets, and toxic components of PF in inducing liver injury, which, however, have differences compared with the actual conditions in clinical practice, and there are still some potential hepatotoxic components and targets of PF that have not been discovered. With the continuous progress in systems biology, establishing the drug-induced liver injury model and the liver injury prediction model based on network toxicology can reduce the cost of animal experiments, improve the toxicity prediction efficiency, and provide new tools for predicting toxic components and targets. To systematically explain the characteristics of liver injury in the application of PF and explore the potential hepatotoxic components and targets of PF, we reviewed the related articles published by China National Knowledge Infrastructure (CNKI), Wanfang Data, VIP, and PubMed from 1962 to 2021 and analyzed the characteristics and influencing factors of liver injury caused by PF in the patients. Furthermore, we summarized the chemical components of PF and the components entering blood. By reviewing the mechanism, targets, and components of PF in inducing liver injury that were discovered by in vivo and in vitro experiments, we summarized the known compounds in PF that may cause liver injury. Finally, the current methods for building the prediction model of PF-induced liver injury were summarized, and the predicted toxic components and targets were introduced. The possible factors of PF in causing liver injury were explained from three aspects: clinical characteristics, preclinical studies, and computer-assisted network prediction, which provide a reference for predicting the risk of PF-induced liver injury.

ABSTRACT

This study was designed to comprehensively characterize and identify the chemical components in traditional Chinese medicine Psoraleae Fructus by establishing an ultra-high performance liquid chromatography/quadrupole time-of-flight mass spectrometry(UHPLC-Q-TOF-MS) method in combination with in-house library. The chromatographic separation conditions(stationary phase, column temperature, mobile phase, and elution gradient) and key MS monitoring parameters(capillary voltage, nozzle voltage, and fragmentor) were sequentially optimized via single-factor experiments. A BEH C_(18) column(2.1 mm×100 mm, 1.7 μm) was finally adopted, with the mobile phase consisting of 0.1% formic acid in water(A) and acetonitrile(B) at the flow rate of 0.4 mL·min~(-1) and column temperature of 30 ℃. Auto MS/MS was utilized for data acquisition in both positive and negative ion modes. By comparison with reference compounds, analysis of the MS~2 fragments, in-house library retrieval and literature research, 83 compounds were identified or tentatively characterized from Psoraleae Fructus, including 58 flavonoids, 11 coumarins, 4 terpenoid phenols, and 10 others. Sixteen of them were identified by comparison with reference compounds, and ten compounds may have not been reported from Psoraleae Fructus. This study achieved a rapid qualitative analysis on the chemical components in Psoraleae Fructus, which provided useful reference for elucidating its material basis and promoting the quality control.

ABSTRACT

With the rise of incidence, fatality rate, and number of young cases, diabetes mellitus has been one of the seven major diseases threatening human health. Although many antidiabetic drugs(oral or for injection) are available, the majority have serious side effects during the long-term use. Thus, it is of particularly vital to develop new drugs with low risk and definite effect. Psoraleae Fructus, a traditional medicinal widely used in the folk, has hypoglycemic, anti-osteoporosis, antitumor, estrogen-like, and anti-inflammatory effects. Thus, it has great clinical application potential. Chinese medicine and the active ingredients, characterized by multiple targets, multiple pathways, and multiple effects in the treatment of diabetes mellitus, have distinct advantages in clinical application. However, the safety of Chinese medicine remains to be a challenge, and one of keys is to clarifying the mechanism of a single Chinese medicinal and its active ingredients. With the method of literature research, this study summarized and analyzed the hypoglycemic mechanisms of Psoraleae Fructus and its main active ingredients over the last decade: regulating glucose metabolism, improving insulin resistance, and directly acting on pancreatic β-cells. The result is expected to serve as a reference for further research on the effects of Psoraleae Fructus and its main chemical constituents in lowering blood glucose and preventing diabetes mellitus and the clinical application.

ABSTRACT

An ultra-high performance liquid chromatography-tandem mass spectrometry(UHPLC-MS/MS) method was established to investigate the pharmacokinetic behaviors of psoralenoside, isopsoralenoside, calycosin-7-glucoside, ononin, psoralen, isopsoralen, methylnissolin, and neobavaisoflavone in rat plasma after oral administration of Bufei Huoxue Capsules. After SD rats were administered with Bufei Huoxue Capsules suspension by gavage, blood samples were collected from the inner canthus at different time points. After protein precipitation, plasma samples were separated on ACQUITY UPLC BEH C_(18) column(2.1 mm×100 mm, 1.7 μm). The mobile phase consisted of acetonitrile(A) and water(B) containing 0.1% formic acid in gradient elution. The positive and negative ions were measured simultaneously in the multi-reaction monitoring(MRM) mode. The pharmacokinetic parameters were calculated and fitted by DAS 3.2.8. Psoralenoside, isopsoralenoside, calycosin-7-glucoside, ononin, psoralen, isopsoralen, methylnissolin, and neobavaisoflavone were detected in the rat plasma after drug administration, with AUC_(0-t) of(3 357±1 348),(3 555±1 696),(3.03±0.88),(2.21±0.33),(1 787±522),(2 295±539),(5.69±1.41) and(3.40±0.75) μg·L~(-1)·h, and T_(max) of(1.56±0.62),(1.40±0.70),(0.21±0.05),(0.25±0.12),(0.26±0.11),(0.34±0.29),(0.74±0.59), and 0.25 h. The method is proved specific and repeatable and is suitable for the determination of psoralenoside, isopsoralenoside, calycosin-7-glucoside, ononin, pso-ralen, isopsoralen, methylnissolin, and neobavaisoflavone in the rat plasma, which can be applied to pharmacokinetic study.

ABSTRACT

This study was designed to explore the alleviating effect and mechanism of Glycyrrhizae Radix et Rhizoma against Psora-leae Fructus-induced liver injury based on network pharmacology and cell experiments. The active components of Glycyrrhizae Radix et Rhizoma and Psoraleae Fructus were first retrieved from the Encyclopedia of Traditional Chinese Medicine(ETCM), Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP), Comparative Toxicogenomics Database(CTD), and literature and further screened by SwissADME. The obtained 25 potential toxic components of Psoraleae Fructus and 29 flavonoids in Glycyrrhizae Radix et Rhizoma were input into the SwissTargetPrediction for target predication. A total of 818 targets related to liver injury were screened out based on GeneCards and MalaCards, and 91 common targets of Psoraleae Fructus, Glycyrrhizae Radix et Rhizoma, and liver injury were obtained from Venny. STRING was applied for constructing the PPI network, and Metascape for analyzing the biological processes and signaling pathways that common targets participated in. Cytoscape was used to construct the component-target-disease network and component-target-pathway network for Glycyrrhizae Radix et Rhizoma against Psoraleae Fructus-induced liver injury. The predicted core targets were proto-oncogene tyrosine-protein kinase(SRC), phosphatidylinositol 4,5-bisphosphate 3-kinase subunit alpha(PIK3 CA), RAC-alpha serine/threonine-protein kinase(AKT1), etc, with PI3 K-AKT signaling pathway, MAPK signaling pathway, apoptosis, Toll-like receptor signaling pathway, and NF-κB signaling pathway mainly involved. Following the scree-ning of the main toxic and pharmacodynamic components, the pharmacodynamic effects were investigated by cell experiments. The results showed that licochalcone A was mainly responsible for alleviating coryfolin-induced liver injury, licochalcone B for coryfolin-and psoralidin-induced liver injury, and echinatin for corylifolinin-and bakuchiol-induced liver injury. The preliminary revealing of the alleviating effect of Glycyrrhizae Radix et Rhizoma on Psoraleae Fructus-induced liver injury and the prediction of related mechanisms will provide reference for further mechanism research and reasonable clinical compatibility.

ABSTRACT

Psoraleae Fructus (PF) is a well-known traditional herbal medicine in China, and it is widely used for osteoporosis, vitiligo, and other diseases in clinical settings. However, liver injury caused by PF and its preparations has been frequently reported in recent years. Our previous studies have demonstrated that PF could cause idiosyncratic drug-induced liver injury (IDILI), but the mechanism underlying its hepatotoxicity remains unclear. This paper reports that bavachin isolated from PF enhances the specific stimuli-induced activation of the NLRP3 inflammasome and leads to hepatotoxicity. Bavachin boosts the secretion of IL-1β and caspase-1 caused by ATP or nigericin but not those induced by poly(I:C), monosodium urate crystal, or intracellular lipopolysaccharide. Bavachin does not affect AIM2 or NLRC4 inflammasome activation. Mechanistically, bavachin specifically increases the production of nigericin-induced mitochondrial reactive oxygen species among the most important upstream events in the activation of the NLRP3 inflammasome. Bavachin increases the levels of aspartate transaminase and alanine aminotransferase in serum and hepatocyte injury accompanied by the secretion of IL-1β via a mouse model of lipopolysaccharide-mediated susceptibility to IDILI. These results suggest that bavachin specifically enhances the ATP- or nigericin-induced activation of the NLRP3 inflammasome. Bavachin also potentially contributes to PF-induced idiosyncratic hepatotoxicity. Moreover, bavachin and PF should be evaded among patients with diseases linked to the ATP- or nigericin-mediated activation of the NLRP3 inflammasome, which may be a dangerous factor for liver injury.

ABSTRACT

Coumarins are the main active components in Psoraleae Fructus. To study the multi-component pharmacokinetics of Psoraleae Fructus, this study established a sensitive and rapid ultra-pressure liquid chromatography coupled to tandem mass spectrometry(UPLC-MS/MS) method for simultaneous determination of psoralen, isopsoralen, psoralenoside, and isopsoralenoside in rat plasma. After validation, the method was applied to the investigation of pharmacokinetics of psoralen, isopsoralen, psoralenoside, and isopso-ralenoside in rats after single and multiple administration of Psoraleae Fructus extract. The results revealed that the exposure of psoralen and isopsoralen in rat plasma was high after a single intragastric administration of Psoraleae Fructus extract, with an AUC_(0-∞) of 443 619-582 680 and 167 314-276 903 ng·mL~(-1)·h~(-1), respectively. Compared with these two compounds, the exposure of psoralenoside and isopsoralenoside was lower with marked gender difference. After 7-day administration of Psoraleae Fructus extract to rats, the AUC_(0-∞) of psoralen and isopsoralen was 29 701-81 783 and 39 234-89 914 ng·mL~(-1)·h~(-1), respectively, which was significantly lower than that at the first day(P<0.05), and that of psoralenoside and isopsoralenoside was 7 360-19 342 and 8 823-45 501 ng·mL~(-1)·h~(-1), respectively. There was no significant gender difference in exposure of psoralenoside and isopsoralenoside in male and female rats. However, the exposure of psoralenoside and isopsoralenoside in male rats was reduced(P<0.05), and the t_(1/2) and mean residence time(MRT) were shortened, suggesting that the removal of these two compounds from the body was accelerated.

ABSTRACT

An immunologically stressed rat model was used in a metabolomics study on the ability of Paeoniae Rubra Radix to reduce the liver toxicity of Psoraleae Fructus. Different groups of rats were given the extracts of Psoraleae Fructus and Psoraleae Fructus together with Paeoniae Rubra Radix or combined with a non-toxic dose of lipopolysaccharide (LPS). The biochemical indices of liver function and pathological changes in liver tissue were used to evaluate histopathological changes. UHPLC-QTOF/MS was used to analyze the metabolic profile of serum samples, combined with principal component analysis (PCA) and orthogonal partial least squares discriminant analysis (OPLS-DA) methods. The HMDB database and Metabo Analyst online tool were used for biomarker identification and metabolic pathway-enrichment analysis. The results show that the co-treatment Psoraleae Fructus and LPS resulted in significant liver injury, indicated by the elevation of aspartate aminotransferase (AST) and alanine aminotransferase (ALT) activities, as well as obvious pathological changes. Liver injury was significantly decreased by treatment with Paeoniae Rubra Radix. Metabolomic analysis showed that the addition of Paeoniae Rubra Radix ameliorated the abnormal serum metabolism in rats mainly through regulation of arachidonic acid metabolism and glycerophospholipid metabolism pathways.

Xianling Gubao is a common and effective medicine in the treatment of orthopedic diseases. In recent years, it has been reported to be associated with liver injury. However, through the analysis of the adverse drug reaction reports and key hospital cases, we found that there is considerable incomplete information in the reports of Xianling Gubao-related liver injury cases retrieved from the literature. Thus, it is difficult to accurately judge causality between the drug and liver injury. Six cases of liver injury related to Xianling Gubao were identified in key hospitals, two of which achieved the clinical diagnosis according to the assessment of the integrated evidence chain method. We further analyzed the public health data of all residents in Yinzhou. The gross incidence rate of Xianling Gubao-related liver injury was 0.034%, which corresponds to a level of rare incidence. This revealed that Xianling Gubao-related liver injury has significant divergence in individuals and an idiosyncratic nature. The gross incidence of liver injury related to Xianling Gubao was lower than that of other medicines for the treatment of orthopedic diseases. Based on the idiosyncratic drug-induced liver injury model mediated by immune stress, it was found that Epimedii Folium and Psoraleae Fructus were the major components that lead to liver injury, and the liver injury caused by a full prescription was less serious than that encountered with only Epimedii Folium and Psoraleae Fructus. This suggests that the other 4 herbs (Dipsaci Radix, Anemarrhenae Rhizoma, Rehmanniae Radix,Salviae Miltiorrhizae Radix et Rhizoma) can prevent/alleviate the liver injury. Through disassembled prescription analysis, we found that the attenuation efficacy of Salviae Miltiorrhizae Radix et Rhizoma was the most significant. In conclusion, Xianling Gubao may cause idiosyncratic liver injury in a tiny minority of susceptible individuals, but the incidence risk is lower than that of other commonly used drugs for orthopedic disease. Xianling Gubao should be discreetly applied to patients with immune stress. The major components that induced liver injury in Xianling Gubao were Epimedii Folium and Psoraleae Fructus, and Salviae Miltiorrhizae Radix et Rhizoma appears to attenuate this toxicity. This study provides a reference for the rational clinical medication with Xianling Gubao.

OBJECTIVE@#To analyze clinical feature and information of medication to explore the risk signals of preparations containing Psoraleae Fructus (BGZP) related with hepatobiliary adverse drug reactions (ADR), in order to reinforce pharmacovigilance.@*METHODS@#A retrospective study was conducted based on hepatobiliary ADR related with BGZP from the China Adverse Drug Reaction Monitoring System in years from January 2012 to December 2016. Serious and general ADRs were analyzed and assessed.@*RESULTS@#There were 355 cases of hepatobiliary ADR related to BGZP. Both the amount of cases and the proportion of serious ADR showed an increasing growth by years (P<0.05). It was found that 10.43% of 355 cases may be involved with irrational drug use, including overdose, repeated medication, and combination of multiple drugs. There were 190 cases which used BGZP (non-combination), and they were mainly for common in diseases caused by abnormal immune activation (accounting for 40.53% of the total cases). Especially at the age group with the most cases with age of 41-50 years, the cases associated with immunological diseases of female were obviously more than that of male (P<0.05). The latency of hepatobiliary ADR related to BGZP ranged from 1 to 386 days, and the median latency was 27.5 days, along with the range of cumulative dose (0.45-520.02 g) as well as the daily dose (0.09-2.64 g/d) after the conversion.@*CONCLUSIONS@#Cases of hepatobiliary ADR related to BGZP showed significant individual differences, and there was no correlation between drug usage duration and dosage and the occurrence of hepatobiliary ADR. It may be similar with idiosyncratic drug-induced liver injury, and recommended that BGZP should be used with more caution under monitoring liver function, especially in female patients with immunological diseases.

ABSTRACT

An UPLC-MS/MS method for rapid and simultaneous determination of psoralen, isopsoralen, apigenin, genistein, bavaisoflavone, neobavaisoflavone, bavachin, bavachinin, psoralenoside, and isopsoralenoside of Psoraleae Fructus in beagle dog plasma was established, and then the method was applied in the pharmacokinetic study after oral administration of Psoraleae Fructus extract to beagle dogs. The pharmacokinetic parameters were calculated by the software of WinNonlin. A Waters HSS-T3 column(2.1 mm×100 mm,1.8 μm)was used for liquid chromatography separation with acetonitrile-water(containing 0.004% formic acid) as the mobile phase for gradient elution.The mass spectrometry was detected using electrospray ion source(ESI) under multi-reaction monitoring mode(MRM), as well as positive ion mode. Analysis time only takes 8.5 min. The methodological study in terms of specificity, accuracy, precision, linear range, recovery, matrix effect, and stability, was validated. The LC-MS analysis method established in this experiment was simple, specific, accurate, reliable, and meet the requirement of pharmacokinetic study in plasma after administration of Psoraleae Fructus extract to beagle dogs. Six beagle dogs received intragastric administration of Psoraleae Fructus extract, T_(max) of 10 chemical components is 1.92-5.67 h; among them, C_(max) of psoralen, isopsoralen, psoralenoside and isopsoralenoside is 383-3 613 ng·mL~(-1), and AUC_(0-∞) is 3 556-18 949 ng·h·mL~(-1), t_(1/2) is 2.45-4.83 h. C_(max) of the remaining six compounds is 0.81-19.9 ng·mL~(-1), AUC_(0-∞ )is 6.54-178 ng·h·mL~(-1), t_(1/2) is 2.95-7.29 h. The UPLC-MS/MS analysis method established in this study was proved to be accurate and sensitive that it can be applied to the pharmacokinetic study of beagle dogs after oral administration of Psoraleae Fructus extract.

ABSTRACT

Psoraleae Fructus is the dried and mature fruit of the legume Psoralea corylifolia. It is warm in nature, pungent and bitter in flavor, and attributive to the kidney and spleen meridians. Its main effect include warming the kidney and assisting Yang, absorbing Qi and relieving asthma, warming the spleen and relieving diarrhea, etc., and it also can for external use of eliminating wind and freckle. Clinically, Psoraleae Fructus is mainly used for the treatment of impotence due to kidney deficiency, soreness of waist and knees, vitiligo, etc. The existing studies have shown that Psoraleae Fructus has a variety of pharmacological effect, such as anti-tumor, anti-oxidant, anti-bacterial, anti-inflammatory, promoting bone growth and protecting cardiovascular. But at the same time, many studies at home and abroad have found that taking Psoraleae Fructus and its compounds for a long time or in large doses can cause liver toxicity, phototoxicity, nephrotoxicity, etc. The most common is liver toxicity, most of the clinical reports on the toxicity of psoralen are caused by drug-induced liver injury events, which limits the clinical use of Psoraleae Fructus and can't exert its proper therapeutic effect. Therefore, it is particularly important to fully understand the toxicological mechanism of liver injury caused by Psoraleae Fructus and its attenuation methods. In this paper, by consulting the domestic and foreign related literatures in recent years that reported the hepatotoxicity of Psoraleae Fructus, the four aspects of clinical report on liver injury, hepatotoxic components, toxicological mechanisms and attenuation methods of Psoraleae Fructus were reviewed, including bile acid stasis and oxidative stress. The hepatotoxicity of Psoraleae Fructus was discussed in terms of reaction, mitochondrial damage, liver fat deformation, etc., and the attenuation methods of Psoraleae Fructus were summarized from the aspects of compatibility attenuation and processing attenuation, aiming to comprehensively and objectively clarify Psoraleae Fructus. The potential toxicological mechanism of lipid-induced hepatotoxicity and research progress in attenuation were expected to provide a theoretical basis for further study of Psoraleae Fructus hepatotoxicity and clinical rational use of drugs.

Objective: To study bakuchiol and its derivatives of cyclohexane soluble part in 70% ethanol aqueous extract of Psoraleae Fructus and their inhibition on nitric oxide (NO) production in lipopolysaccharides (LPS)-activated murine macrophage RAW 264.7 cell lines. Methods: The compounds were separated and purified by silica gel column and high performance liquid chromatographies, and their structures were determined by spectroscopic data analyses. Using LPS-activated RAW 264.7 cell line models in vitro, all of the isolated compounds were evaluated for the inhibition against NO production. Results: Twelve compounds were obtained and identified as bakuchiol (1), 12,13-dihydro-12,13-epoxybakuchiol (2), Δ3,2-hydroxylbakuchiol (3), 12-oxobakuchiol (4), psoracorylifol B (5), psoracorylifol C (6), (12’S)-bisbakuchiol C (7), Δ1,3-bakuchiol (8), 13-methoxyisobakuchiol (9), bisbakuchiol B (10), bisbakuchiol A (11), and 12,13-dihydro-12,13-dihydroxybakuchiol (12), respectively. For the inhibition of NO production in the LPS-activated RAW 264.7 cell line model, a positive inhibitor, L-N6-(1-iminoethyl)-lysine (L-NIL), was used and showed the half maximal inhibitory concentration (IC50) value of (10.29 ± 1.10) μmol/L. The IC50 values of the assayed compounds 1, 3, 5, 10 and 11 were all more than 50 μmol/L, compounds 8, 9 and 12 were comparable to that of L-NIL, whereas the IC50 values of compounds 2, 4 and 7 were less than that of the positive inhibitor with statistically significance. Conclusion: Compound 4 is a new natural product. The results of the bioactivity assays indicated that compounds 2, 4, 7, 8, 9 and 12 are potential anti-inflammatory agents.

ABSTRACT

Recently, the hepatotoxicity issue regarding to Psoraleae Fructus (PF) has attracted remarkable concerns, which highlights the urgent need to explore the toxicity attenuation method for PF. In this study, we proposed an alcohol soaking and water rinsing method for pre-processing PF based on the record in the classics - "Lei Gong Pao Zhi Lun", aiming to attenuate the potential hepatotoxicity of PF. The optimal pre-processing methods and parameters were investigated by U*12(108) uniform design coupled with 3D-cultured human-derived liver organoids model and high-content imaging. The results showed that there were significant variations among the hepatotoxicity intensities of different pre-processed PF products. Four factors, including the concentration of alcohol, the ratio of material and alcohol in alcohol soaking, the time of alcohol soaking and the times of water rinsing, were found as independent significant factors (all P<0.01). The optimal pre-process parameters were further predicted and verified as follows: the alcohol concentration is 80%, the times of alcohol soaking is 3, the ratio of alcohol and material of alcohol soaking is 3, the time for alcohol soaking is 30 h, the ratio of water and material of water rinsing is 2, the times of water rinsing is 3, the time water rinsing is 12 h and the time of steaming is 5 h. This research demonstrated that the alcohol soaking and water rinsing method can effectively reduce the potential hepatotoxicity of PF. This method provides a reference for reducing the risk of PF liver injury from the perspective of Chinese medicinal materials pre-processing.

ABSTRACT

Objective:Arrange long-term toxicity experiments by a uniform design method, so as to explore the effect of different extracts of Psoraleae Fructus on liver toxicity in rats and mice, and find the drug factors that cause psoralen liver toxicity. Method:Based on the factors of processing, extraction technology, dosage and treatment course, each experimental group was arranged by uniform design method. A total of 220 SD rats and 220 Kunming mice with half male and half female were divided into normal groups and drug groups 1 to 8. The corresponding drugs (50% alcohol extract of salt Psoraleae Fructus in rats 2.57 g·kg-1, mice 5.14 g·kg-1, 95% alcohol extract of Psoraleae Fructus in rats 0.51 g·kg-1, mice 1.02 g·kg-1, 70% alcohol extract of salt Psoraleae Fructus in rats 1.71 g·kg-1, mice 3.42 g·kg-1, water extract of Psoraleae Fructus in rats 1.03 g·kg-1, mice 2.06 g·kg-1, water extract of salt Psoraleae Fructus in rats 1.03 g·kg-1, mice 2.06 g·kg-1, 70% alcohol extract of Psoraleae Fructus in rats 1.71 g·kg-1, mice 3.42 g·kg-1, 95% alcohol extract of salt Psoraleae Fructus in rats 0.51 g·kg-1, mice 1.02 g·kg-1, 50% alcohol extract of Psoraleae Fructus in rats 2.57 g·kg-1, mice 5.14 g·kg-1) were administered by gavage daily. The body weight and food intake of the rats and mice were measured once a week. After the treatment course, the rats were anesthetized with sodium pentobarbital, and blood was taken from the abdominal aorta, and the mice were sacrificed by removing the eyeballs, and the liver and brain were taken to calculate the organ coefficients. Serum was taken to determine liver function-related indicators, and the liver was taken for histopathological examination by hematoxylin-eosin (HE) staining. Result:The liver visceral-brain ratio of female rats in group 3 were significantly increased (P<0.05). The liver quality, visceral-body ratio and visceral-brain ratio of male mice in groups 1 to 3 were significantly increased (P<0.05, P<0.01). Histopathological manifestations in mice were more obvious than those in rats. Histopathology showed hepatocyte hypertrophy in the central area of liver lobules in mice, in particular in group 3. According to the multiple regression equation, there were interactions between extraction technology, processing, dosage and treatment course, and the extraction technology was positively correlated with the pathological score of liver injury. Based on the results of visual analysis and other indicators, it is concluded that the extraction technology factor is most relevant to psoralen liver toxicity of Psoraleae Fructus. Conclusion:Psoraleae Fructus has the hepatotoxicity, which is related to ethanol extraction technology; alcohol extraction is more toxic than water extraction, and 70% ethanol extraction is the most toxic. Besides, there are species differences, with a more significant hepatotoxicity in mice than that in rats.

ABSTRACT

Objective: The purpose of this study was to establish a QAMS analytical method for 16 compounds in Psoraleae Fructus, and attempt to evaluate the quality difference among different batches of Psoralen Fructus by chemometrics. Methods: All experiments were performed on three different HPLC instruments. Isopsoralen was used as the internal reference substance to determine the relative correction factors of the other 15 compounds. The robustness and durability of the measured relative correction factors of the 15 compounds were evaluated on different chromatograph instruments and columns; And the measurement result deviation was compared between QAMS method and the external standard method. Results: Under the established chromatographic conditions, the relative correction factors of 15 compounds in Psoraleae Fructus had high accuracy, good durability, and good reproducibility under different experimental conditions. The results obtained from two analysis methods showed no significant deviation. The results obtained by the new established QAMS analytical method showed that the consistency of compound types among different batches of Psoraleae Fructus were better, but the content of different componounds was relatively different. Conclusion: The newly established QAMS analytical method for simultaneous determination of 16 compounds in Psoraleae Fructus provides a more efficient method to evaluate the comprehensive quality of Psoraleae Fructus from different sources.

Objective: To investigate the effects of Buguzhi Pills and three kinds of simulative Buguzhi Pills on diarrhea rats with spleen-kidney yang deficiency, and to confirm the rationality of the salt-processed composition of Buguzhi Pills. Methods: SD rats were randomly divided into nine groups: normal group, model group, positive drug group, salt-processed Psoraleae ructus group, raw Psoraleae Fructus group, Buguzhi Pills (salt-processed Psoraleae Fructus + salt-processed Foeniculi Fructus) group and three kinds of simulative Buguzhi Pills groups (salt-processed Psoraleae Fructus + raw Foeniculi Fructus; raw Psoraleae Fructus + salt-processed Foeniculi Fructus; raw Psoraleae Fructus + raw Foeniculi Fructus). A complex method of hydrocortisone and sennae folium was adopted to establish the diarrhea model due to spleen-kidney yang deficiency. After administration, the organ indexes of thymus, spleen, kidney, adrenal gland, and testis were measured; The pathological changes of corresponding organs were observed; The serum levels of motilin and testosterone were detected by ELISA. Results: Compared with the normal group, the food intake and body weight reduced; The organ indexes decreased in varying degrees; The above organs showed obvious pathological changes under the microscope; the level of motilin showed an upward trend, and the level of testosterone decreased significantly in model group. The above symptoms and indicators were improved in varying degrees after administration, and the effect of Buguzhi Pills was better than that of Psoraleae Fructus alone and three kinds of simulative Buguzhi Pills on the whole. Conclusion :The efficacy of Buguzhi Pills on warming kidney-spleen yang and relieving diarrhea was superior to that of Psoraleae Fructus alone, and the antidiarrheal effect can be enhanced by salt-processing.

Psoraleae Fructus (Buguzhi) is dried mature fruit of Psoralea corylifolia. It is spicy and bitter in favor and warm in nature, and distributed to kidney and spleen meridians. Psoralea Fructus has the effects of warming kidney and tonifying Yang,absorbing Qi and relieving asthma,warming spleen and stopping diarrhea; for external use,it can remove beverage. It is mainly used to treat impotence and nocturnal emission cold and knee pain,kidney deficiency and asthma caused by kidney Yang deficiency; for external use, it can treat vitiligo and alopecia areata. Its principle chemical components are coumarins,monoterpene phenols,flavonoids,with an estrogen-like effect as well as anti-inflammatory,antibacterial,antifungal,anti-oxidation,anti-tumor and immunomodulatory activities, and can be used for the prevention and treatment of osteoporosis,bacterial infections,asthma and osteoarthritis. However,the long-term administration with large doses of psoralen have the potential risk of causing liver damage,with phototoxicity,nephrotoxicity and reproductive toxicity,which restricts its application in clinical therapy. Therefore,it is essential to fully understand the chemical composition and mechanism of psoralen. Through the review and summarization of literatures about psoralen at home and abroad,this article systematically reviews the chemical constituents and toxicity of Psoralea Fructus, provides Chinese and English names,structural formulas,molecular formulas,molecular weights,CAS numbers of the compounds and the toxicity information about psoralen reported in recent years, in order to provide a reference for further study on the compositional spectrum and pharmacokinetics,toxic kinetic studies and clinical medication safety of psoralen.

ABSTRACT

The traditional Chinese medicine standard decoction is prepared on the basis of the theory of traditional Chinese medicine and clinical application. With reference to the modern extraction method,the single decoction of traditional Chinese medicine is prepared by the standardized process,and the establishment of its quality standards is conducive to standardizing clinical medication. This research is to set an evaluation standard for the quality of salt-processed Psoraleae Fructus standard decoction. Twelve batches of salt-processed Psoraleae Fructus standard decoctions were prepared. The contents of psoralen and isopsoralen were determined,the transfer and extract rates were calculated,and the pH value was measured; HPLC fingerprint method was established for analysis. The results of the 12 batches of samples revealed that the transfer rates of psoralen and isopsoralen were 17. 10%-26. 40%,14. 70%-22. 70%,respectively; the extract rate was between 14. 7%-27. 0%,and the pH value was between 5. 4-6. 9. Moreover,7 common chromatographic peaks were determined based on fingerprint by using similarity evaluation system for chromatographic fingerprint of traditional Chinese medicine( 2012 A).The similarities of the 12 batches of samples were analyzed and compared,and the results showed that the similarities were all higher than0. 9. In this study,the preparation method for salt-processed Psoraleae Fructus decoction was standard,with high similarities in fingerprint. This study build a convenient and reliable method of comprehensive quality evaluation,with a high precision,stability and repeatability,which can provide a reference for the quality control of salt-processed Psoraleae Fructus dispensing granules.

ABSTRACT

Objective To establish a quality evaluation system for standard decoction of salt-processed Psoraleae Fructus (PF). Methods Fifteen batches of crude drugs were collected and processed into decoction pieces with salt complied with the 2015 Edition of the Chinese Pharmacopoeia, and then the standard decoctions of salt-processed PF were prepared as freeze-dried powders. Based on the established HPLC fingerprint, seven common peaks were recognized and the main chemical constituents were identified in combination with time-of-flight mass spectrometry. Results Benzofuran glycosides and furanocoumarins turned out to be the main composition of standard decoction of salt-processed PF. The dry extract yielding rate varied from 16.31% to 20.59%, while the total contents of psoralen and isopsoralen were 1.17%—1.50% with a transfer rate ranging from 13.55% to 23.57%, and the fingerprint similarity of 15 batches of samples were all higher than 0.9. Conclusion This stable and reliable method of comprehensive quality evaluation for standard decoction of salt-processed PF may provide reference for quality control of salt-processed PF dispensing granules and related preparations.
